Depends how much your foot is in the throttle.

Sport will be the worst, as it holds gears and keeps rpms up. Manual is really up to how ypu drive it and Drive, should give you the best mileage.

I've been conducting an experiment. I spent a little over 350 miles strictly in Sport mode the entire time. I got exactly 18.5 mpg in what I call Suburban driving. I was not driving in my traditional aggressive mode so this is probably the best I could get. I have since spent another 350+ miles in strictly the Drive mode, again, same mild, suburban driving pattern. I averaged exactly 21.5 mpg for that phase.

I'm now finishing (about 320+ miles) the last phase in what will likely become my actual daily driver mode. Again, suburban driving 95% in Drive but with consistent moves to Sport mode for any turn-ins at intersections or turns into slightly steep climes. The goal is to avoid, completely, any perceived loss of momentum while waiting for the transmission to shift down one or two gears, sometimes three, before I get to a moderate acceleration to maintain full momentum through the turn. I do, of course, find myself rolling on the accelerator just a bit because it is fun but I keep it far from my "full-on" push. In this mode i'm clearly averaging 21.2 mpg.

I've been keeping my driving, overall, in the mild mode because I recently got two tickets. I've since installed a radar detector, am using Waze and will soon apply Veil (for Lasers) when the weather cools a bit. I will likely maintain this "Daily Driving" style with a slight increase in roll-on for fun which will likely push mileage down to around 20-20.5 mpg.

I've not done any extended highway driving yet but when I do it will likely be at 80-90 mph so I'll attempt to capture a realistic mpg number for that speed. I'm sure the current advertised mpg for around 70 mph highway speeds is fairly accurate.

Having done all that, I really don't care about mileage...I was just curious. What I love about the S5 is the power and the flexibility of the transmission to accommodate different driving styles without giving up much of anything. I'm over my "Manual" bigotry of the last 25 years. Modern Tiptronic trannies with a minimum of 350+ hp is now my cup of tea and I love Audi's version thereof.

Around the city in bumper to bumper traffic in individual mode (auto engine, dynamic sound and steering, idle stop on) I get around 13-14mpg. I was able to take it about 800 miles to and from my parents house and I averaged 31 mpg in individual going between 70-80 on the highway (minimal driving off the highway). When I am in dynamic mode around the city, it is more of 10-12 and on the highway 25.

S5 MPG in different drive modes

The gas mileage sucks in this car not that I care, but my Stage 2 B8 S4 got better gas mileage

This tank is driving in Dynamic mode with a mix of S and D, ac on all the time, 60/40 city/highway driving

This tank was comfort mode, ac on less than 50% of the time, 60/40 highway city, always watching the boost gauge trying NOT to be in boost, never flooring it, basically the most boring way to drive a car like a old grandma lol.
